APC2(Adenomatous Polyposis Coli 2)是APC基因家族的成员之一,与著名的肿瘤抑制基因APC(APC1)同源。APC基因家族的主要共性是参与Wnt信号通路的调控,通过促进β-catenin的降解来抑制细胞过度增殖,从而维持组织稳态。APC2主要在神经系统中高表达,尤其在发育中的大脑皮层和海马区,其蛋白产物通过与微管结合参与细胞骨架的组装和神经元迁移,对大脑发育至关重要。APC2还能与APC1协同作用,通过结合轴蛋白(Axin)和糖原合成酶激酶3β(GSK3β)形成“破坏复合体”,促使β-catenin磷酸化并降解,从而阻断Wnt信号通路的异常激活。若APC2发生功能丧失性突变,可能导致β-catenin积累,引发Wnt信号过度活跃,与神经发育障碍如自闭症谱系障碍(ASD)和智力障碍相关。此外,APC2突变还与某些癌症(如胶质瘤)的发生有关,但相比APC1在结直肠癌中的明确作用,APC2的致癌机制尚不清晰。当APC2过表达时,可能过度抑制Wnt通路,影响神经元的正常分化和突触可塑性;而表达降低则可能导致神经迁移缺陷或异常细胞增殖。研究还发现APC2与精神分裂症风险基因DISC1存在相互作用,共同调节神经元形态发生。尽管APC2与APC1功能部分重叠,但APC2在神经系统中的特异性作用使其成为神经发育和退行性疾病研究的重要靶点。
