ARAP2(ArfGAP with RhoGAP domain, ankyrin repeat and PH domain 2)是一种多功能信号调节蛋白,属于ARAP基因家族(包括ARAP1-3)。该家族共性在于同时具备ArfGAP(ADP-核糖基化因子GTP酶激活蛋白)和RhoGAP(Rho家族GTP酶激活蛋白)双重结构域,能通过水解GTP负调控小G蛋白(如Arf和Rho家族)的活性,从而参与细胞骨架重组、囊泡运输和细胞迁移等过程。ARAP2的典型结构包含N端ArfGAP域、中央RhoGAP域、C端PH(pleckstrin homology,可结合磷脂)域和ANK(ankyrin repeat,介导蛋白互作)重复序列。其表达产物通过调控Arf6和RhoA等小G蛋白的活性,在维持内皮细胞屏障功能、血管生成和胰岛素信号转导中起关键作用。主要作用位点包括细胞膜(通过PH域定位)和高尔基体区域。突变可能导致ARAP2的GAP活性丧失,例如错义突变R836Q会破坏RhoGAP功能,引发细胞迁移异常,与转移性乳腺癌相关;而ArfGAP结构域的突变可能干扰GLUT4囊泡转运,促进胰岛素抵抗。该基因与多种疾病关联:过表达会增强RhoA抑制,导致血管过度稳定而阻碍肿瘤血管正常化,但可能改善糖尿病中的血管通透性;低表达则使Arf6持续活化,促进乳腺癌侵袭转移。在基因家族中,ARAP2区别于ARAP1/3的特点是其特异性响应PI(3,4,5)P3磷脂信号,且对Arf6的调控更强。当ARAP2过表达时,会过度抑制RhoA通路,减弱应力纤维形成,影响细胞收缩;而表达不足时,Arf6介导的内吞作用亢进,导致EGFR等受体酪氨酸激酶降解受阻,可能促进肿瘤生长。目前该基因的中文译名"含RhoGAP结构域、锚蛋白重复序列和PH结构域的ArfGAP蛋白2"虽为直译但未被广泛采用,文献中多沿用ARAP2原名。
