BCRP1(乳腺癌耐药蛋白1,也称为ABCG2)属于ATP结合盒(ABC)转运蛋白超家族,是一个重要的跨膜转运蛋白,主要功能是参与多种内源性和外源性物质的主动外排,包括药物、毒素和代谢产物。它在肠道、肝脏、血脑屏障和胎盘等组织中高表达,起到保护机体免受有害物质侵害的作用。BCRP1的特点是其广泛的底物特异性,能够识别并转运多种结构不同的化合物,如抗癌药物(如甲氨蝶呤、拓扑替康)、抗生素和类黄酮等。突变可能影响其转运功能,导致药物敏感性改变或毒性积累。例如,某些突变(如Q141K)会降低蛋白表达或功能,影响药物代谢,增加某些化疗药物的毒性风险。BCRP1过表达与多药耐药(MDR)相关,尤其在癌症治疗中,可能导致化疗失败;而表达降低可能增加药物在体内的积累,提高疗效但同时也可能增加副作用。BCRP1属于ABC转运蛋白家族,该家族成员均利用ATP水解产生的能量进行底物跨膜转运,参与多种生理过程如营养吸收、脂质代谢和细胞解毒。此外,BCRP1与某些疾病密切相关,如高尿酸血症和痛风(因其参与尿酸排泄),以及某些遗传性疾病如新生儿黄疸(因胆红素转运异常)。研究BCRP1对优化药物治疗和开发新型抑制剂以克服耐药性具有重要意义。
