CA2(碳酸酐酶2,Carbonic Anhydrase II)是一种由CA2基因编码的锌金属酶,属于碳酸酐酶(Carbonic Anhydrase, CA)基因家族。该家族共有16个成员(CA1-CA15及CA相关蛋白),共性是通过催化二氧化碳与水可逆反应生成碳酸氢盐和质子(CO2 + H2O ⇌ HCO3− + H+),参与酸碱平衡、离子转运及代谢调控。CA2是家族中活性最高的同工酶,广泛分布于红细胞、肾脏、胃黏膜、骨骼及中枢神经系统(尤其是胶质细胞),主要作用位点为细胞质。其生物学功能包括:1)调节体液pH值,在红细胞中协助二氧化碳运输;2)在肾脏近端小管促进HCO3−重吸收;3)在胃壁细胞参与胃酸分泌;4)在骨组织中影响破骨细胞功能,调控骨吸收。CA2突变可导致常染色体隐性遗传病——碳酸酐酶2缺乏综合征(Marble brain disease),表现为肾小管酸中毒、骨质疏松及脑钙化,因突变使酶活性丧失,破坏酸碱平衡和骨代谢。该基因与青光眼(房水分泌异常)、癫痫(神经pH紊乱)及某些癌症(如肾细胞癌)相关。CA2过表达可能加剧肿瘤微环境酸化(原英文:tumor acidosis),促进侵袭转移;而表达降低会引发代谢性酸中毒或骨质异常。CA2还通过调节HCO3−水平影响其他基因,如下调可能抑制SLC4家族(HCO3−转运蛋白)功能,过表达则激活Wnt/β-catenin通路(原英文:Wnt/β-catenin signaling)加速骨修复。其活性可被磺胺类药物(如乙酰唑胺)特异性抑制,用于治疗青光眼和高原病。
