DNAJB11(也称为Hsp40或J-domain蛋白家族成员)属于热休克蛋白40(Hsp40/DNAJ)家族,该家族以共同含有保守的J结构域(J-domain)为特征,能够与热休克蛋白70(Hsp70)协同作用,参与蛋白质折叠、转运、降解等过程,维持细胞稳态。DNAJB11主要位于内质网(ER),作为分子伴侣辅助Hsp70调控新生肽链的正确折叠,防止错误折叠蛋白聚集,并在内质网相关降解(ERAD)途径中发挥作用。其突变可能导致内质网应激(ER stress)加剧,与多种疾病相关,如多囊肾病(ADPKD)和某些神经退行性疾病。若DNAJB11过表达,可能增强错误折叠蛋白的清除能力,但过度激活可能干扰正常蛋白稳态;若表达降低,则错误折叠蛋白积累,触发未折叠蛋白反应(UPR),长期可导致细胞凋亡。该基因家族(DNAJ/Hsp40)成员均通过J结构域激活Hsp70的ATP酶活性,但不同亚型具有组织特异性或功能分工,如部分成员参与线粒体或细胞质蛋白质量控制。DNAJB11的病理突变可能破坏其与Hsp70的相互作用,影响内质网功能,进而引发疾病表型。
