ELAVL2(也称为HuB)属于ELAVL(胚胎致死异常视觉样)基因家族,该家族包括四个成员:ELAVL1(HuR)、ELAVL2(HuB)、ELAVL3(HuC)和ELAVL4(HuD)。这些基因编码RNA结合蛋白,主要功能是通过结合mRNA的AU富集元件(ARE)来调控基因表达的转录后水平,影响mRNA的稳定性、翻译效率和定位。ELAVL2主要在神经元中表达,尤其在发育中的神经系统和成人脑组织中高度活跃,对神经元的发育、可塑性和功能维持至关重要。它通过调控靶mRNA(如神经营养因子和突触相关蛋白的mRNA)的稳定性,参与神经分化、突触形成和记忆形成等过程。ELAVL2的突变或表达异常可能导致神经系统疾病,例如神经退行性疾病或神经发育障碍。研究表明,ELAVL2的过表达可能增强神经元的存活和突触可塑性,但也可能与某些神经系统肿瘤的发生相关。相反,ELAVL2表达降低可能导致神经元功能受损,影响认知能力或增加神经退行性病变的风险。ELAVL家族成员的共性在于它们都含有三个RNA识别模体(RRM),能够特异性结合ARE序列,但各成员的组织分布和功能侧重有所不同。ELAVL2在神经系统中具有独特作用,其异常表达或功能失调可能通过扰乱RNA代谢网络而影响其他基因的表达,进而干扰神经系统的正常功能。
