ELK4(ETS transcription factor ELK4)属于ETS转录因子家族,该家族成员共享一个保守的DNA结合域(ETS结构域),能够识别并结合特定的DNA序列(GGAA/T核心基序),调控下游基因的转录。ELK4主要通过与血清反应因子(SRF)形成复合物,结合到血清反应元件(SRE)上,参与细胞增殖、分化、凋亡和应激反应的调控。它在MAPK信号通路中起关键作用,可被ERK、JNK等激酶磷酸化激活,进而调控如c-Fos等早期反应基因的表达。ELK4的突变或异常表达可能影响其DNA结合能力或转录活性,导致细胞周期紊乱或肿瘤发生。研究发现,ELK4在某些癌症(如前列腺癌、乳腺癌)中过表达,可能通过促进细胞增殖和抑制凋亡参与肿瘤进展;而敲低ELK4可能抑制肿瘤生长,但同时也可能影响正常细胞的应激响应。此外,ELK4与家族成员ELK1、ELK3(NET/SAP2)功能部分冗余,但各自在组织分布和靶基因选择上存在差异。例如,ELK4在睾丸中高表达,可能与生殖细胞发育相关。该基因的异常还可能与心血管疾病或神经退行性疾病有关,具体机制仍需进一步研究。
