ENO1(Enolase 1)是一种编码α-烯醇化酶的基因,属于烯醇化酶基因家族。该家族成员(包括ENO1、ENO2和ENO3)在糖酵解途径中催化2-磷酸甘油酸转化为磷酸烯醇式丙酮酸,是能量代谢的关键酶。ENO1基因家族的共性在于它们均具有高度保守的烯醇化酶结构域,并在不同组织中特异性表达:ENO1广泛表达于多种组织,ENO2主要在神经元和神经内分泌细胞中表达,而ENO3则特异性存在于肌肉组织中。ENO1除了参与糖酵解外,还具有多种非经典功能,如作为热休克蛋白在应激条件下保护细胞,以及作为纤溶酶原受体参与细胞迁移和侵袭过程。ENO1在细胞质和细胞膜上均有分布,其膜结合形式与肿瘤转移密切相关。ENO1突变可能导致酶活性丧失,影响细胞能量代谢,并与某些遗传性代谢疾病相关。研究发现ENO1过表达与多种癌症(如肺癌、乳腺癌和胶质瘤)的进展和不良预后显著相关,其机制涉及促进肿瘤细胞增殖、迁移和侵袭。此外,ENO1过表达还可通过激活PI3K/AKT等信号通路促进肿瘤发生。相反,ENO1表达降低会导致细胞能量代谢障碍,影响细胞生长和存活。ENO1还被发现是某些自身免疫疾病(如类风湿性关节炎)的自身抗原,其抗体水平与疾病活动度相关。在感染性疾病中,某些病原体可利用ENO1作为受体入侵宿主细胞。ENO1的表达受缺氧诱导因子HIF-1α调控,在缺氧条件下表达上调。由于其在肿瘤发生发展中的重要作用,ENO1已成为潜在的癌症治疗靶点,针对ENO1的小分子抑制剂和抗体药物正在研发中。
