FLT1(Fms相关酪氨酸激酶1)是一种编码血管内皮生长因子受体1(VEGFR-1)的基因,属于VEGFR家族。该家族还包括FLK1(VEGFR-2)和FLT4(VEGFR-3),它们共同参与调控血管生成和淋巴管生成。FLT1主要通过与血管内皮生长因子(VEGF-A、VEGF-B)和胎盘生长因子(PlGF)结合来发挥作用,但其激酶活性较弱,主要作为诱饵受体调节VEGF信号通路的可用性,从而影响血管形成和维持血管稳态。FLT1在胚胎发育、伤口愈合和女性生殖系统(如胎盘形成)中起关键作用。FLT1的突变或表达异常与多种疾病相关,例如子痫前期患者中可溶性FLT1(sFLT1)水平升高会抑制VEGF信号,导致血管功能障碍和高血压。此外,FLT1在肿瘤血管生成中也有重要作用,其过表达可能促进肿瘤生长和转移,而降低表达则可能抑制血管生成,但同时也可能影响正常血管功能。FLT1基因家族成员均含有免疫球蛋白样结构域和酪氨酸激酶结构域,通过调控细胞增殖、迁移和存活来影响血管及淋巴管系统的发育和功能。
