HDHD5-AS1 is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the human chromosome 1p36.33 region, serving as a regulatory counterpart to the neighboring HDHD5 gene, which encodes a member of the haloacid dehalogenase-like hydrolase domain (HDHD) family known for its role in phosphate ester bond hydrolysis. Although HDHD5-AS1 does not encode a protein, it likely exerts biological influence through mechanisms typical of lncRNAs, such as epigenetic modulation, transcriptional interference, or acting as a microRNA sponge to fine-tune the expression of HDHD5 and other targets within metabolic and signaling networks. Emerging evidence suggests that HDHD5-AS1 may function as a tumor suppressor, as its expression is notably downregulated in colorectal cancer tissues, implying that its loss could disrupt the regulation of cell proliferation and metabolic pathways, whereas its overexpression might restore or alter these homeostatic controls. While the specific molecular interactions and downstream effects of HDHD5-AS1 mutations remain under investigation, the gene’s role in maintaining cellular function is consistent with the broader hydrolytic activities of the HDHD family, and further research is required to fully elucidate its precise mechanistic contributions and clinical relevance in disease contexts.
