MAPK8(丝裂原活化蛋白激酶8)也称为JNK1(c-Jun N端激酶1),属于MAPK(丝裂原活化蛋白激酶)家族中的JNK亚家族。该基因编码的蛋白是一种丝氨酸/苏氨酸激酶,在细胞应激反应、炎症、凋亡、增殖和分化等过程中发挥关键作用。MAPK8主要通过磷酸化转录因子(如c-Jun、ATF2)调控基因表达,从而影响细胞对压力、紫外线辐射或细胞因子的反应。其活性受上游激酶MKK4和MKK7的调控。MAPK8基因突变可能导致激酶活性异常,与多种疾病相关,包括神经退行性疾病(如阿尔茨海默病)、炎症性疾病和癌症。过表达MAPK8会增强细胞凋亡和炎症反应,可能促进肿瘤发生或神经细胞死亡;而表达降低则可能削弱细胞对压力的应对能力,影响发育和免疫反应。MAPK家族成员均参与信号转导,通过三级激酶级联(MAPKKK-MAPKK-MAPK)传递外界刺激至细胞核。JNK亚家族(包括MAPK8、MAPK9、MAPK10)主要响应应激信号,具有相似的结构域和激活机制。研究表明,MAPK8在肝脏代谢、胰岛素抵抗和肥胖中也起调节作用,其抑制剂正在被研究作为潜在的治疗药物。
