MicroRNAs (miRNAs) are short (20–24 nucleotides) noncoding RNAs that regulate gene expression post-transcriptionally by affecting mRNA stability and translation in multicellular organisms. miRNAs are transcribed by RNA polymerase II as capped and polyadenylated primary transcripts (pri-miRNAs), which may be either protein-coding or noncoding. The primary transcript is cleaved by the ribonuclease III enzyme Drosha to produce an approximately 70-nucleotide stem-loop precursor miRNA (pre-miRNA), which is further processed by cytoplasmic Dicer ribonuclease to yield mature miRNA and antisense miRNA star (*miRNA) products. Mature miRNAs are incorporated into RNA-induced silencing complexes (RISCs), which recognize target mRNAs through imperfect base pairing with the miRNA and most often cause translational repression or destabilization of the target mRNA.
