MicroRNAs (miRNAs) are short (20–24 nucleotide) noncoding RNAs that post-transcriptionally regulate gene expression in multicellular organisms by affecting mRNA stability and translation. They are transcribed by RNA polymerase II as capped and polyadenylated primary transcripts (pri-miRNAs), which may be protein-coding or noncoding. Drosha ribonuclease III cleaves the primary transcript to produce an approximately 70-nucleotide stem-loop precursor miRNA (pre-miRNA), which cytoplasmic Dicer ribonuclease further processes into mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into the RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing and most often causes translational repression or destabilization of the target mRNA.
