MicroRNAs (miRNAs) are short (20–24 nt) noncoding RNAs that post-transcriptionally regulate gene expression in multicellular organisms by affecting mRNA stability and translation. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs), which can be either protein-coding or noncoding. The primary transcript is cleaved by Drosha ribonuclease III to yield an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which cytoplasmic Dicer further cleaves to produce the mature miRNA and antisense miRNA star (miRNA*) product. The mature miRNA is incorporated into the RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly leads to translational repression or destabilization of the target mRNA.
