MicroRNAs (miRNAs) are short (20–24 nucleotides) noncoding RNAs that post-transcriptionally regulate gene expression in multicellular organisms by affecting mRNA stability and translation. miRNA genes are transcribed by RNA polymerase II as capped and polyadenylated primary transcripts (pri-miRNAs), which can be protein-coding or noncoding. The primary transcript is cleaved by the ribonuclease III enzyme Drosha to generate an approximately 70-nucleotide stem-loop precursor miRNA (pre-miRNA), which is further cut by cytoplasmic Dicer to produce mature miRNA and antisense miRNA star (miRNA*) products. Mature miRNA is incorporated into the RNA-induced silencing complex (RISC), which recognizes target mRNAs by imperfect base pairing and most often leads to translational repression or destabilization of the target mRNA.
