MicroRNAs (miRNAs) are short (20–24 nt) noncoding RNAs that post-transcriptionally regulate gene expression in multicellular organisms by affecting mRNA stability and translation. They are transcribed by RNA polymerase II as capped and polyadenylated primary transcripts (pri-miRNAs), which may be protein-coding or noncoding. Pri-miRNAs are cleaved by Drosha ribonuclease III to generate ~70-nt stem-loop precursor miRNAs (pre-miRNAs), which are further processed by cytoplasmic Dicer into mature miRNAs and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into the RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing and most often leads to translational inhibition or destabilization of the target mRNA.
