MicroRNAs (miRNAs) are short (20–24 nt) noncoding RNAs that post-transcriptionally regulate gene expression in multicellular organisms by affecting mRNA stability and translation. They are transcribed by RNA polymerase II as parts of capped and polyadenylated primary transcripts (pri-miRNAs), which may be protein-coding or noncoding. The pri-miRNA is cleaved by the RNase III enzyme Drosha to produce an approximately 70-nucleotide stem-loop precursor miRNA (pre-miRNA), which cytoplasmic Dicer further cleaves to yield mature miRNA and antisense miRNA star (miRNA*) products. Mature miRNA is incorporated into the RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing and most commonly causes translational inhibition or destabilization of the target mRNA.
