MicroRNAs (miRNAs) are short (20–24 nucleotide) noncoding RNAs that post-transcriptionally regulate gene expression in multicellular organisms by affecting mRNA stability and translation. They are transcribed by RNA polymerase II as parts of capped and polyadenylated primary transcripts (pri-miRNAs), which can be protein-coding or noncoding. Pri-miRNAs are cleaved by Drosha RNase III to generate approximately 70-nucleotide stem-loop pre-miRNAs, which are further processed by cytoplasmic Dicer into mature miRNAs and antisense miRNA* products. Mature miRNAs are loaded into the RNA-induced silencing complex (RISC), where imperfect base pairing with target mRNAs typically causes translational repression or destabilization.
