RALA(RAS-like proto-oncogene A)属于RAS超家族中的RAL小G蛋白家族,这个家族还包括RALB基因。RAL家族成员作为分子开关,在GTP结合(激活态)和GDP结合(失活态)间转换,调控细胞信号传导。RALA主要参与囊泡运输、细胞迁移、增殖和肿瘤发生等过程,其效应蛋白包括RALBP1(RAL结合蛋白1)和EXOC(外泌体复合体),通过调控内吞作用和胞吐作用影响细胞膜动力学。RALA在多种组织中表达,尤其在脑、肾脏和胎盘中活性较高。RALA突变可能破坏其GTP酶活性,导致持续激活状态,与多种癌症(如膀胱癌、肺癌和胰腺癌)的发展相关。过表达RALA会增强肿瘤细胞的侵袭性和转移能力,促进EMT(上皮-间质转化),同时激活PI3K/AKT和MAPK等促生存通路;而敲低RALA则抑制肿瘤生长并增加化疗敏感性。RALA与RALB功能部分冗余,但RALA更偏向调控细胞增殖,RALB则更多参与免疫应答和细胞存活。RAL家族共性包括:依赖GEF(鸟苷酸交换因子)激活和GAP(GTP酶激活蛋白)失活,通过C端脂修饰锚定细胞膜,并参与膜运输和细胞骨架重组。RALA还与神经退行性疾病(如阿尔茨海默病)相关,可能通过影响APP(淀粉样前体蛋白)的运输加剧β-淀粉样蛋白积累。此外,RALA通过调控CDC42和RAC1等Rho家族蛋白影响细胞极性。药物开发中,靶向RALA-GTP结合位点的抑制剂(如erastin衍生物)正在研究中,用于阻断肿瘤转移。
