RHOA(Ras homolog family member A)属于Rho GTPase家族,是小G蛋白超家族的重要成员,在细胞骨架重组、细胞迁移、增殖和基因表达调控等过程中发挥核心作用。RHOA通过GTP/GDP结合状态切换来调控下游信号通路,激活状态(GTP结合)可结合效应蛋白如ROCK(Rho-associated kinase)和mDia(哺乳动物Diaphanous相关蛋白),进而调节肌动蛋白应力纤维形成、细胞黏附和收缩。其主要作用位点包括细胞膜和胞质,通过与其他Rho家族成员(如RAC1、CDC42)协同调控细胞极性。RHOA突变(如G17V、L57V)可能导致组成性激活或功能丧失,与多种疾病相关:激活突变促进肿瘤侵袭转移(如胃癌、乳腺癌),而功能缺失突变可能影响心血管发育(如动脉导管未闭)。RHOA过表达会增强细胞迁移能力但抑制细胞周期进程,可能导致纤维化疾病或肿瘤转移;表达降低则破坏细胞连接(如上皮-间质转化受阻),影响胚胎发育(如神经嵴细胞迁移异常)。该基因与弥漫性大B细胞淋巴瘤(DLBCL)关系密切,约20%病例存在RHOA p.D120N突变。RHOA属于Rho基因家族(含20多个成员),家族共性为:均含GTP酶结构域,通过GTP水解循环调控细胞形态和运动;共享GEF(鸟苷酸交换因子)/GAP(GTP酶激活蛋白)调控系统;参与整合生长因子和黏附信号。在免疫系统中,RHOA还调控T细胞受体信号转导,其表达异常可能导致自身免疫疾病。最新研究发现RHOA在机械信号转导中起核心作用,能感知细胞外基质硬度并调控YAP/TAZ通路。药物开发中,针对RHOA-ROCK通路的抑制剂(如法舒地尔)已用于治疗脑血管痉挛和肺动脉高压。
