RPL15(核糖体蛋白L15)是核糖体大亚基的组成蛋白之一,属于RPL基因家族,该家族编码核糖体60S亚基的结构蛋白,共同参与蛋白质合成的翻译过程。RPL15在核糖体组装和稳定中起关键作用,其表达产物与rRNA结合,帮助维持核糖体结构完整性,并影响翻译的准确性和效率。主要作用位点是细胞质中的核糖体,但部分研究表明它可能参与核内pre-rRNA加工。若RPL15发生功能丧失性突变(如无义突变或移码突变),可能导致核糖体生物发生缺陷,引发核糖体病(ribosomopathy),典型表现为 Diamond-Blackfan贫血(DBA),这是一种以红细胞生成障碍为特征的遗传性疾病,患者常伴有发育异常和癌症易感性。RPL15的异常表达还与多种肿瘤相关,例如在肝癌和乳腺癌中过表达可能通过激活mTOR信号通路促进细胞增殖,而低表达可能导致核糖体应激(ribosomal stress)并触发p53介导的细胞周期阻滞或凋亡。在基因家族层面,RPL家族成员均含有保守的核糖体蛋白结构域,多数具有碱性氨基酸富集区以结合RNA,且普遍存在冗余补偿机制——单个成员表达下调时其他家族成员可能部分代偿其功能。需要注意的是,RPL15的过表达可能干扰核糖体平衡,导致翻译全局速率改变或特定mRNA(如癌基因)的选择性翻译增强;而表达不足则可能引发未折叠蛋白反应(UPR)或自噬等应激途径。目前"核糖体病"等术语的中文译法存在争议,部分文献保留英文原词ribosomopathy。

核糖体,催化蛋白质合成的细胞器,由一个小40S亚基和60S大亚基。一起这些亚基组成的4 RNA种类和大约80结构不同的蛋白质。该基因编码一种核糖体蛋白,它是60S亚基的一个组成部分。该蛋白属于L15E家族核糖体的蛋白质。它位于细胞质中。这个基因股序列相似与酵母核糖体蛋白YL10基因。虽然这个基因被称为RPL10,其官方符号是RPL15.本基因已表明在某些食管肿瘤中过表达相对于正常匹配的组织。选择性剪接结果在多个抄本变形。作为典型编码核糖体蛋白的基因,有该基因通过基因组分散的多个经处理的假基因。 [由RefSeq的,2011年11月提供]
