RPL36(核糖体蛋白L36)是核糖体60S大亚基的组成蛋白之一,属于RPL基因家族(核糖体蛋白家族),该家族成员均为核糖体结构蛋白,共同参与核糖体组装及蛋白质翻译过程。RPL36在胞质中与rRNA结合,协助维持核糖体结构稳定性,并直接参与mRNA解码和肽键形成等翻译核心功能。其突变可能导致核糖体组装异常,引发核糖体病(ribosomopathy),典型表现为造血障碍(如Diamond-Blackfan贫血)、骨骼发育异常或癌症易感性增加。RPL36过表达可能破坏核糖体亚基平衡,导致翻译效率下降或错误蛋白积累,与某些肿瘤(如结直肠癌)的进展相关;而低表达则可能因全局蛋白质合成减少引发细胞凋亡,尤其在快速增殖组织中影响显著。该基因家族成员均具有高度保守性,多数为小分子量碱性蛋白,其功能冗余性使单个基因突变常表现为组织特异性表型。目前研究发现RPL36在肝癌组织中表达异常,可能通过调控特定癌基因翻译影响肿瘤发生,但具体机制仍需进一步研究。专业术语解释:核糖体(ribosome)是细胞中合成蛋白质的分子机器,由rRNA和蛋白质组成;翻译(translation)指以mRNA为模板合成蛋白质的过程;核糖体病(ribosomopathy)指因核糖体功能缺陷导致的遗传性疾病。
