RPS7(核糖体蛋白S7)是核糖体小亚基的重要组成部分,属于RPS基因家族(核糖体蛋白家族),该家族成员普遍参与核糖体组装和蛋白质翻译过程。RPS7在细胞质中与rRNA结合形成40S核糖体亚基,直接参与mRNA解码和蛋白质合成,其核心功能是维持核糖体结构稳定性并调控翻译效率。该基因在几乎所有组织中均有基础表达,尤其在增殖活跃的细胞(如干细胞、肿瘤细胞)中表达较高。突变可能导致核糖体组装异常,引发核糖体病(ribosomopathy),典型表现为Diamond-Blackfan贫血(先天性纯红细胞再生障碍性贫血),患者会出现造血功能障碍、骨骼畸形等。RPS7还通过结合MDM2抑制p53降解,间接参与肿瘤抑制调控。过表达可能增强蛋白质合成能力促进细胞增殖,与肝癌、乳腺癌等恶性肿瘤进展相关;而低表达会导致翻译效率下降,引发细胞周期阻滞或凋亡。该基因家族(RPS系列)成员均具有高度保守的碱性氨基酸结构域,能与rRNA静电结合,但各成员在核糖体上的空间定位和具体调控机制存在差异。近期研究发现RPS7可通过可变剪接产生异构体,可能影响特定mRNA的翻译选择性。在病毒感染过程中,某些病毒会劫持RPS7以促进自身蛋白合成,例如丙型肝炎病毒(HCV)的IRES序列就依赖RPS7进行病毒蛋白翻译起始。
