S100A8(也称为钙粒蛋白A或MRP8)是一种属于S100蛋白家族的基因,其编码的蛋白质与S100A9形成异源二聚体(两个不同蛋白质结合),共同参与炎症和免疫调节。S100蛋白家族的共性是其成员均含有EF手型结构域(一种能结合钙离子的特殊蛋白结构),依赖钙离子调节功能,并在细胞增殖、分化及炎症反应中发挥作用。S100A8主要在髓系细胞(如中性粒细胞和单核细胞)中表达,其蛋白产物具有促炎和抗菌活性,通过结合Toll样受体4(TLR4)或晚期糖基化终末产物受体(RAGE)激活免疫信号通路。突变或异常表达可能导致功能紊乱,例如S100A8/A9复合物过度释放会加剧慢性炎症(如类风湿性关节炎和炎症性肠病),而某些突变可能削弱其抗菌能力。过表达时,S100A8会促进炎症因子(如IL-6、TNF-α)释放,导致组织损伤或癌症进展(如乳腺癌和结直肠癌);低表达则可能削弱先天免疫防御。该基因还与自身免疫性疾病(如系统性红斑狼疮)和心血管疾病相关,其水平可作为炎症标志物。在肿瘤微环境中,S100A8既能抑制免疫细胞活性,又能促进肿瘤转移,呈现双重作用。
