RAB17是RAB基因家族(Ras-related in brain,Ras相关脑蛋白家族)的成员之一,属于小GTP酶超家族。RAB家族主要调控细胞内囊泡运输、膜 trafficking(膜转运)和信号转导等过程。RAB17在多种组织中表达,但尤其在极化上皮细胞(如肾脏和肠道)中发挥重要作用,参与顶端 recycling endosome(循环内体)的运输和蛋白质分选,影响细胞极性建立和维持。其表达产物RAB17蛋白通过GTP/GDP结合状态切换(类似分子开关)调控囊泡与特定膜区室的对接与融合。突变可能导致RAB17功能丧失或异常激活,破坏囊泡运输,进而影响细胞极性相关疾病(如某些癌症或肾脏疾病)。研究发现RAB17过表达可能促进某些肿瘤(如乳腺癌)的转移,因其能增强侵袭相关蛋白的运输;而敲低表达则可能损害上皮屏障功能,导致极性紊乱。该基因与溶酶体储存障碍、纤毛病等也存在潜在关联。RAB家族共性包括:均含有保守的GTP酶结构域,通过结合GTP(激活态)或GDP(失活态)循环发挥作用,并需要GEF(鸟苷酸交换因子)和GAP(GTP酶激活蛋白)调控;多数C端带有异戊二烯化修饰(脂修饰)以保证膜定位。目前中文常将"recycling endosome"直译为循环内体,但更准确的译法应为再循环内体(英文原词)。
