RPL30(核糖体蛋白L30)是核糖体60S大亚基的组成蛋白之一,属于RPL基因家族(核糖体蛋白家族),该家族成员均参与核糖体组装及蛋白质翻译过程。RPL30在细胞质中与rRNA结合,协助维持核糖体结构稳定并催化肽键形成,其核心功能是促进mRNA的翻译效率,影响细胞增殖、分化等基础生命活动。该基因在几乎所有组织中广泛表达,尤其在代谢活跃的细胞(如肝细胞、肿瘤细胞)中表达较高。RPL30若发生功能丧失性突变(如无义突变或移码突变),可能导致核糖体组装缺陷,引发核糖体病(ribosomopathy),典型表现为 Diamond-Blackfan贫血(骨髓衰竭伴红细胞生成障碍)或发育异常;而错义突变可能仅导致翻译效率下降,引发细胞应激反应。研究发现,RPL30在多种癌症(如乳腺癌、结直肠癌)中过表达,其机制可能与促进肿瘤细胞快速增殖所需的蛋白质合成有关;相反,若表达量降低则可能触发p53依赖的细胞周期阻滞。RPL基因家族的共性包括:高度保守的序列特征、与rRNA的相互作用域、以及多数成员在应激条件下可脱离核糖体转而调控特定mRNA的翻译(这一功能称为"核糖体异质性" ribosome heterogeneity)。需要注意的是,RPL30的小鼠同源基因(Rpl30)还被发现可通过结合自身mRNA的5'UTR来负反馈调节其翻译水平,这种自体调控机制在核糖体蛋白中较为常见。
