RPL12(核糖体蛋白L12)是核糖体大亚基的重要组成部分,属于RPL基因家族(核糖体蛋白家族),该家族成员普遍参与核糖体组装和蛋白质翻译过程。RPL12在核糖体60S亚基中发挥结构支撑作用,并与翻译延伸因子eEF-2相互作用促进肽链延伸。其表达产物具有典型的碱性蛋白特性,含有高度保守的N端结构域和C端α螺旋结构。该基因的主要作用位点是细胞质中的核糖体,突变可能导致核糖体组装异常,引发翻译效率下降或错误,进而影响细胞增殖。研究发现RPL12突变与戴蒙德-黑凡贫血症(Diamond-Blackfan anemia,一种核糖体病)相关,其单倍剂量不足(haploinsufficiency)会导致红系祖细胞分化障碍。过表达RPL12可能通过增强翻译活性促进肿瘤发生,已在乳腺癌和肝癌中发现其表达上调现象;而表达降低则会导致核糖体应激(ribosomal stress)并激活p53通路,引发细胞周期阻滞。RPL基因家族的共性包括:编码的蛋白均参与核糖体生物发生(ribosome biogenesis),多数含有RNA结合域,在进化上高度保守,且常呈现多拷贝基因特征。需要注意的是,RPL12在部分文献中也被称为RPL23(旧命名),其功能冗余性可能被同家族成员部分补偿。最新研究还发现RPL12具有核糖体之外的功能,如参与DNA损伤修复和细胞凋亡调控。
