RPS19(核糖体蛋白S19)是核糖体小亚基的组成蛋白之一,属于RPS基因家族(核糖体蛋白家族),该家族成员均参与核糖体组装及蛋白质翻译过程。RPS19在40S核糖体亚基中发挥作用,直接参与mRNA解码和蛋白质合成,其功能对细胞增殖、分化及稳态维持至关重要。若RPS19发生突变(如错义突变或缺失),可能导致核糖体生物合成障碍,引发 Diamond-Blackfan贫血(DBA),这是一种以红细胞生成衰竭为特征的遗传性疾病,患者表现为严重贫血、先天性畸形及癌症易感性。RPS19突变占DBA病例的约25%,突变会破坏其与18S rRNA的结合能力,导致核糖体组装异常。RPS19过表达可能干扰核糖体亚基平衡,引发p53依赖的细胞周期阻滞(核糖体应激反应),而表达降低则直接损害蛋白质合成效率,影响造血干细胞功能。此外,RPS19通过调控VEGF等因子参与血管生成,其异常表达可能与肿瘤进展相关。RPS基因家族的共性包括:高度保守性、在翻译中起结构性或催化作用、部分成员(如RPS19)具有核糖体外的调控功能(如参与细胞凋亡或信号转导)。术语解释:核糖体(ribosome)是细胞内合成蛋白质的分子机器,由rRNA和核糖体蛋白组成;mRNA(信使RNA)携带遗传信息指导蛋白质合成;VEGF(血管内皮生长因子)促进血管形成。
