Intention-treated water inhibits migration of human glioblastoma cells via downregulation of motility-associated proteins.

Building on a previously reported finding that intention-treated water reduced human glioblastoma cell migration, this study had two aims: to replicate the earlier observation using a more physiologically precise migration assay, and to identify candidate molecular mechanisms by profiling key migration-regulatory proteins. In a double-blind, controlled protocol, a single batch of ultrapure water was exposed to focused mental intention by four Buddhist monks; a paired control batch received no treatment. U-87MG glioblastoma cells were cultured in media prepared from either the intention-treated or control water. Cell migration was quantified using a Transwell assay across three technical replicates, and the expression of eleven migration-regulatory proteins was assessed by Western blot across three replicates from the same water batches. Across all three assay runs, cells cultured in intention-treated media consistently exhibited reduced migration relative to controls (p < 0.002, one-sample t-test against normalized control mean). Western blot analysis showed consistent downregulation of VASP and RhoB across runs (p < 0.004 and p < 0.0003, respectively, after correction for multiple analyses), with eight additional factors trending in the same direction. Within a single double-blind treatment occasion, intention-treated water was associated with a reproducible reduction in glioblastoma cell migration and consistent suppression of the actin cytoskeletal regulatory axis. This study involved repeated testing of a single batch of treated vs. control water, rather than repeatedly testing the intentional treatments. Future studies using independently treated water batches will be required before claims about the generality of the intentional effect can be made.
