ENO2(Enolase 2)是一种编码γ-烯醇化酶的基因,属于烯醇化酶基因家族。该家族包括ENO1、ENO2和ENO3等成员,它们均编码烯醇化酶的同工酶,参与糖酵解途径,催化2-磷酸甘油酸转化为磷酸烯醇式丙酮酸。ENO2主要在神经元和神经内分泌细胞中高表达,因此也被称为神经元特异性烯醇化酶(NSE)。ENO2的生物学功能不仅限于能量代谢,还涉及神经发育、突触可塑性以及细胞存活。其表达产物γ-烯醇化酶在神经系统中具有重要作用,可作为神经损伤或神经内分泌肿瘤的生物标志物。ENO2突变可能导致酶活性降低,影响糖酵解效率,进而干扰神经元能量供应,与某些神经系统疾病如神经退行性疾病或脑缺血相关。此外,ENO2过表达常见于小细胞肺癌、神经母细胞瘤等肿瘤中,可能与肿瘤细胞的 Warburg 效应(有氧糖酵解增强)有关,促进肿瘤生长和侵袭。降低ENO2表达则可能影响神经元功能,导致认知障碍或神经发育异常。烯醇化酶基因家族的共性在于它们均参与糖代谢,但不同成员具有组织特异性表达模式,例如ENO1广泛表达于多种组织,ENO3主要在肌肉中表达。ENO2因其神经元特异性而成为研究神经疾病和肿瘤的重要靶点。
