FGR基因(也称为FGR原癌基因或SRC2)属于SRC家族酪氨酸激酶基因家族,该家族成员在细胞信号传导中起关键作用,参与调控细胞生长、分化、迁移和存活等过程。FGR编码一种非受体型酪氨酸激酶,主要表达于造血细胞(如B细胞、巨噬细胞和中性粒细胞)中,通过磷酸化下游靶蛋白传递信号。FGR蛋白包含SH3、SH2和激酶结构域,能与其他信号分子相互作用形成复合物。FGR的功能异常与多种疾病相关,其过度激活可能导致炎症反应增强和自身免疫疾病,而功能丧失则可能影响免疫细胞功能。研究发现FGR在某些白血病(如慢性淋巴细胞白血病)中表达升高,可能促进癌细胞存活和增殖。FGR基因突变可能破坏其激酶活性或蛋白相互作用能力,导致信号传导异常。在动物模型中,FGR敲除小鼠表现出免疫系统轻微缺陷但对大多数生理功能无严重影响,说明该基因功能可能存在冗余。SRC家族基因(包括SRC、YES、LYN等)的共性是都能通过酪氨酸磷酸化调控下游信号通路,许多成员在癌症中异常激活。FGR过表达可能增强炎症反应并促进某些肿瘤发展,而表达降低可能削弱免疫细胞功能但通常不会引起严重表型。该基因与炎症性疾病和血液系统恶性肿瘤的关联使其成为潜在的治疗靶点,针对SRC家族激酶的抑制剂(如达沙替尼)已用于临床但缺乏对FGR的特异性。
