PIMREG, also known as FAM64A, is a gene encoding a protein that functions as a critical regulator of cell cycle progression, particularly at the G1/S transition, and is a member of the FAM64 gene family, which is characterized by conserved protein domains involved in modulating cell proliferation and transcriptional regulation. The PIMREG protein exerts its effects by interacting with cyclin-dependent kinase (CDK) complexes, thereby facilitating the advancement of the cell cycle; however, its dysregulated overexpression, frequently observed in malignancies such as breast, lung, and colorectal cancers, can drive uncontrolled cellular proliferation and contribute to tumorigenesis. Beyond direct interaction with CDKs, PIMREG influences the broader cell cycle regulatory network by altering the expression of key genes such as CDKN1A and CCND1, which can exacerbate cell cycle disorders and genomic instability, particularly when mutations impair its binding capabilities. Although primarily associated with cancer pathogenesis, PIMREG also plays a role in the immune system, potentially contributing to T-cell activation and proliferation. Given that high expression levels of PIMREG are correlated with poor patient prognosis, it holds significant potential as a biomarker for cancer diagnosis and prognostication, while its role as a promoter of proliferation makes it an attractive target for future therapeutic strategies aimed at inhibiting tumor growth.
