RASGRP2(RAS Guanyl Releasing Protein 2)是一种鸟苷酸交换因子(GEF),属于RASGRP基因家族。该家族共有四个成员(RASGRP1-4),它们通过激活RAS和RAP等小G蛋白参与细胞信号传导,调控细胞增殖、分化和存活等过程。RASGRP2主要表达于血小板和巨核细胞中,其核心功能是激活RAP1蛋白,从而促进血小板活化和血栓形成。该基因的突变可能导致血小板功能异常,例如Glanzmann血小板无力症(一种罕见的出血性疾病),表现为血小板无法正常聚集。RASGRP2过表达会增强血小板反应性,增加血栓风险;而表达降低则可能导致出血倾向。此外,RASGRP2还参与免疫调节,影响T细胞和B细胞的激活。研究发现该基因与心血管疾病、炎症性疾病和某些癌症相关。RASGRP基因家族的共性在于都含有REM(RAS交换因子)和CDC25结构域,能够催化GDP/GTP交换从而激活RAS超家族蛋白。RASGRP2的独特之处在于其对RAP1而非RAS具有更强选择性。该基因的调控异常可能影响多个信号通路(如MAPK/ERK),进而干扰细胞生理功能。
