SUMO1(Small Ubiquitin-like Modifier 1)是一种小泛素样修饰蛋白,属于SUMO基因家族(包括SUMO1、SUMO2、SUMO3和SUMO4)。该家族成员通过共价结合靶蛋白(称为SUMO化修饰,SUMOylation)调控其功能,类似于泛素化(ubiquitination),但不会导致蛋白降解,而是影响蛋白稳定性、亚细胞定位、转录活性或蛋白相互作用。SUMO1在进化中高度保守,主要表达于细胞核,参与DNA损伤修复、细胞周期调控、信号转导和应激反应等过程。其作用位点通常为靶蛋白的赖氨酸残基(如转录因子p53、核孔蛋白RanGAP1)。SUMO1突变可能导致修饰异常,例如SUMO1第93位甘氨酸突变为天冬氨酸(G93D)会削弱其与靶蛋白结合能力,引发神经发育缺陷或心血管疾病。研究发现SUMO1与阿尔茨海默病(淀粉样蛋白沉积异常)、癌症(如抑制肿瘤 suppressor p53的活性)及先天性心脏病(如NOTCH1信号通路失调)相关。SUMO1过表达可能增强靶蛋白的稳定性(如稳定HIF-1α促进肿瘤血管生成),但也会干扰正常SUMO化平衡,导致细胞凋亡异常;而表达降低则可能影响基因组稳定性(如削弱DNA修复蛋白Rad51的功能)。SUMO家族共性包括:C末端具有保守的"GG" motif(用于与靶蛋白结合)、依赖E1/E2/E3酶级联反应进行修饰,并通过SENP去SUMO化酶(SUMO-specific proteases)动态调控。需注意"SUMO化"早期曾被译为"类泛素化",但当前学界统一使用"SUMO化修饰"(原英文:SUMOylation)。
