SYK (spleen tyrosine kinase) is a non-receptor tyrosine kinase belonging to the SYK family, which also includes ZAP-70, and is predominantly expressed in hematopoietic lineages such as B cells, T cells, and macrophages, where it plays a pivotal role in immune receptor signal transduction. Functionally, SYK is recruited to the cytoplasmic tails of activated immune receptors, such as the B-cell receptor (BCR) and Fc receptors, by binding to phosphorylated immunoreceptor tyrosine-based activation motifs (ITAMs); its distinctive dual SH2 domains allow it to simultaneously engage two phosphorylated ITAMs, thereby amplifying downstream signaling cascades including PI3K-AKT, MAPK, and NF-κB pathways that govern immune cell proliferation, differentiation, survival, and inflammatory responses. This signaling activity is spatially concentrated at the immunological synapse, such as the contact site between B cells and antigens, and within inflammatory lesions. Dysregulation of SYK activity has significant pathological implications: gain-of-function mutations or overexpression can lead to excessive immune activation, contributing to autoimmune disorders like rheumatoid arthritis, allergic diseases, and certain hematological malignancies including diffuse large B-cell leukemia, whereas loss-of-function mutations may result in immunodeficiency. Consequently, SYK is an attractive therapeutic target, with inhibitors currently under investigation or used to modulate immune responses in conditions such as rheumatoid arthritis, leveraging the potential of reduced SYK expression to suppress aberrant inflammatory reactions.
