LYN是一种属于Src家族激酶(SFK)的非受体酪氨酸激酶基因,在免疫细胞信号传导中发挥核心作用。该基因位于人类8号染色体(8q12.1),编码的Lyn蛋白具有典型的Src家族结构特征:N端肉豆蔻酰化位点、独特结构域、SH3和SH2蛋白相互作用域以及C端的激酶活性域。作为B细胞受体(BCR)信号通路的关键调节者,Lyn通过磷酸化免疫受体酪氨酸激活基序(ITAM)启动下游信号,同时也能通过磷酸化抑制性受体(如CD22和FcγRIIB)的ITIM基序来负向调控信号。在肥大细胞中,Lyn参与FcεRI受体介导的过敏反应信号传导。该基因突变可导致功能获得或功能丧失,功能获得性突变与某些白血病相关,而功能缺失则与自身免疫疾病如系统性红斑狼疮有关。Lyn表达异常会影响多种细胞过程:过表达可能促进肿瘤发生,特别是在慢性淋巴细胞白血病(CLL)和急性髓系白血病(AML)中观察到其异常激活;而表达降低会导致B细胞过度活化,与自身抗体产生相关。在Src家族中,Lyn与Blk、Fyn等成员共享通过脂修饰锚定在细胞膜内侧的特性,都能通过酪氨酸磷酸化调控下游信号。这个家族的特征是都具有调节细胞增殖、分化、存活和迁移的酪氨酸激酶活性,且其活性受C端抑制性酪氨酸(Y507)和激活环酪氨酸(Y396)磷酸化状态的动态平衡控制。最新研究发现Lyn还参与神经退行性疾病如阿尔茨海默病的病理过程,可能通过调节小胶质细胞活性影响神经炎症。针对Lyn的抑制剂如达沙替尼已被开发用于癌症治疗,但需注意其对正常免疫功能的影响。
