MOS基因属于丝裂原活化蛋白激酶激酶激酶(MAP3K)家族,该家族成员在细胞信号转导中起关键作用,主要参与调控细胞增殖、分化、凋亡和应激反应等过程。MOS基因编码一种丝氨酸/苏氨酸蛋白激酶,是MAPK/ERK信号通路的上游激活因子,在卵母细胞减数分裂中发挥核心作用。其表达产物通过磷酸化并激活MEK1,进而激活ERK1/2,最终调控细胞周期进程。MOS的主要作用位点是生殖细胞,尤其在卵母细胞成熟过程中不可或缺。若MOS发生功能丧失性突变,会导致卵母细胞减数分裂阻滞,造成女性不育;而获得性功能突变则可能引发异常增殖,与某些生殖系统肿瘤相关。研究表明MOS过表达会持续激活ERK通路,导致细胞转化和肿瘤发生,已在卵巢癌、乳腺癌等病例中发现其异常高表达。相反,MOS表达降低会破坏减数分裂纺锤体组装,引发染色体分离错误。该基因与母源效应基因家族相关,这类基因的特点是其mRNA在卵子发生过程中被储存,在受精后翻译为蛋白质调控早期胚胎发育。MOS作为该家族代表,其蛋白在受精后被迅速降解,这种精确的时空调控对胚胎正常发育至关重要。在进化上,MOS基因在脊椎动物中高度保守,但在果蝇等无脊椎动物中不存在,提示其在脊椎动物生殖进化中的特殊地位。
